Blastogenic response of peripheral blood lymphocytes during post hatch periods in chicken

Abstract

Development of T-and B-Iymphocyte proliferation in chicken, during day 1 to 60, was assessed using lymphocyte stimulation test with concanavalinA (con A) and Escherichia Coli-lipopolysaccharide (LPS) as T-and B-cell mitogen, respectively. The optical density (OD) of mitogenstimulated cultures was determined in Spectrophotometer at wavelength of 570 nm.The mean delta OD of con-A stimulated cell cultures was 0.033, 0.241, 0.327, and 0.295 on 1, 20, 40, and 60 days of post hatch development, respectively. At post hatch day 1, 20, 40, and 60, the mean delta OD of LPS stimulated cell cultures was 0.07, 0.147, 0.301, and 0.264. The peak blastogenic response was evident on day 35 for T-cells and on day 25 for B-cells. The T cells have a better blastogenic response than the B cells. The peak mitotic response was noted at the age of 25 days for B cells and 35 days for the T cells of the chicken peripheral blood.
