Isolation and identification of Brucella melitensis from sheep in north Gujarat
Abstract
Isolation and molecular methods were adopted for the diagnosis of brucellosis in sheep. Out of a total 51 samples from sheep with history of abortion, two samples (3.92%) yielded Brucella isolates on Brucella Agar medium from seropositive animals. Both the isolates were presumed to be Brucella on cultural, morphological and biochemical characterization. Confirmation of isolates was carried out by genus-specific primer pairs and B4/B5 and JPF/JPR primer pairs were found to be more sensitive. For species level identification among three species specific primers (Omp31, ORFs A0503 and B. abortus+ IS711), only Omp31 primers pair produced desired amplicon of 720 bp and indicated that sheep were infected with Brucella melitensis.
