The effect of washing with tris buffer and preserving in tris-based extender containing 2.5% egg yolk on the preservation of buck semen at 4°C during different time intervals was investigated. Semen was collected from four crossbred Boer bucks by using an artificial vagina. Semen samples with >70% sperm motility were pooled and divided into two aliquots as the non-washed and washed group. Semen was extended with tris citric acid fructose egg yolk (TCFEY) extender containing 2.5% egg yolk and stored at 4°C up to 72 h. Sperm motility and live sperm were significantly (P<0.05) higher in the non-washed than washed group up to 72 h preservation. Hypo-osmotic swelling test- (HOST) reacted spermatozoa were significantly (P<0.05) higher at 0 h and 24 h and intact acrosome at 0 h and 72 h in non-washed than washed group. It was concluded that 2.5% of egg yolk in tris based extender provided better buck semen quality without washing at 4°C for 72 h.
