Grain legumes cultivated as rainfed and irrigated crop in Gujarat are severely affected by whitefly transmitted Yellow Mosaic Disease (YMD). The viruses causing YMD in northern and southern India have been identified to be Mungbean yellow mosaic India virus (MYMIV) and Mungbean yellow mosaic virus (MYMV) respectively, the bipartite begomoviruses belonging to family Geminiviridae. The present work describes molecular cloning of genomic components of YMV infecting mungbean and cowpea. Attempts were made to clone the genomic components of viruses infecting mungbean and cowpea collected from Anand, Palanpur, Navsari and Rajkot areas of Gujarat, by PCR approach using MYMIV specific primers. However, only partial segments of genome (900 bp and 1.8 kb) were obtained. In order to obtain full-length clone (∼2.7 kb), viral replicative DNA was isolated by density gradient centrifugation. Double stranded replicative forms obtained were cloned at BamHI and Kpn I sites. Two different populations, DNA A and DNA B were found among the recombinant clones, which were further authenticated by sequencing. Restriction of DNA A and DNA B component showed pattern different from MYMIV. Preliminary results of sequence analysis indicated the possibility of virus to be different from MYMIV.
