Abstract

Tomato leaf curl Bangalore virus -AVT1 (ToLCBV-AVT1) and Tomato leaf curl New Delhi virus - AVT1 (ToLCNDVAVT1) are known to be mono- and bi-partite begomoviruses affecting tomato in India associated with DNA β respectively (Sivalingam et al., 2004). Agroinoculation of dimeric constructs of genomic components of these two isolates were done in various combinations. Agroinoculation of Nicotiana benthamiana with DNA A + DNA β of ToLCBVAVT1 or with DNA A + DNA B of ToLCNDV-AVT1 produced distinct leaf curl symptoms, whereas agroinoculation with DNA A and DNA A + DNA β of ToLCNDV-AVT1 resulted in leaf curling, and with DNA A + DNA B + DNA β of ToLCNDV-AVT1 resulted in severe leaf curling. In tomato DNA A and DNA A + DNA β of ToLCNDV-AVT1 produced mild leaf curl symptoms; inoculation of DNA A + DNA B produced less severe leaf curl symptoms than DNA A + DNA B + DNA β inoculated plants. Southern analysis of agroinoculated N. benthamiana plants showed that there was increase in the replication of DNA A and DNA B in the DNA A + DNA B + DNA β inoculated plants compared to DNA A + DNA B inoculated plants. These results indicate that i) in monopartite, ToLCBV-AVT1 DNA b lead to systemic infection, and ii) in bipartite, ToLCNDV-AVT1 DNA β increases disease severity and replication of DNA A and DNA B.
