Abstract

A survey was conducted to record the occurrence of whitefly transmitted geminivirus on Bittergourd in four districts of Tamil Nadu. Bitter gourd plants exhibiting the yellow mosaic symptom was collected and was found that the virus was transmitted very efficiently by the whitefly. BGYMV when inoculated passed on to one plant and three weed species viz., Manihot esculenta and Acalypha indica, Croton sparsiflours, Malvastrum coromandelianum. respectively. The sap of BGYMV infected plant and weeds reacted positively with ACMV polyclonal antibody and monoclonal antibody (SCR 18). Naturally and artificially infected bittergourd and artificially inoculated (BGYMV) plant species, weed species viz Manihot esculenta, Malvastrum coromandelianum, Croton sparsiflorus and Acalypha indica respectively were amplified with degenerate primer for the presence of geminivirus. All the above samples were also amplified with ICMV DNA - A specific primer. In both the cases the amplified product size of all the samples was 560 bp. From the above work, it is concluded that BGYMV is a whitefly transmitted geminivirus and more specifically, it is the Indian cassava mosaic virus.
