Abstract

Rice tungro spherical virus (RTSV) is one of the causal agents of tungro disease of rice. The RTSV, which produced mild symptoms on the plant, can be detected using non-radioactive nucleic acid based detection. Northern blotting using glyoxal/DMSO gel gave a better resolution with less background as compared to the formaldehyde gel. A single RNA species was detected from total RNA extracts at 4 weeks post inoculated (wpi) plants and from RNA extract from purified virion. No RNA was detected from healthy and total RNA extracts of 2 wpi plants. The result showed that the viral RNA could successfully be detected upto a concentration of 0.25 mg for both total RNA extracts from 4 wpi and from total RNA extract from purified virion. Results showed that 0.4% SDS and 0.5XSSC at 55°C in the primary washing gives a better resolution over 0.2% SDS and 0.5XSSC at 55°C.
