Abstract

Six day old seedlings of the susceptible pumpkin (Cucurbita moschata Duch. Ex Poir.) cv. Co. 2 were sprayed upon with antiviral principals (AVPs), viz., 10% aqueous leaf extract of Bougainvillea spectabilis Wild and 0.02% acetyl salicylic acid and the seedlings were inoculated with Pumpkin yellow vein mosaic virus (PYVMV) using viruliferous Bemisia tabaci Gen. Total phenols, orthodihydroxy (OD) phenols and catalase activity were measured at 1, 3 and 5 days after ioculation (DAI).

In response to infection with PYVMV, the total phenol and OD phenol contents of pumpkin seedlings increased substantially and gradually from 1 DAI to 5 DAI. Treatment with the AVPs also caused a significant increase in the total phenols & OD phenols. When AVP-treated seedlings were inoculated with PYVMV, there was further upsurge in the total phenol and OD phenol contents. At 5 DAI, the total phenol content was 48.00 to 62.62 per cent more than that of uninoculated control whereas the OD phenol content was 54.16 to 59.91 per cent mroe than that of uninoculated control.

Inoculation of the pumpkin seedlings with PYVMV resulted in 57.49% increase in the catalase activity at 1 DAI. Thereafter, the catalase activity showed a severe downward trend registering only 15.85% and 15.40% increase over the uninoculated control at 3 DAI and 5 DAI respectively. Both applicaiton of AVPs and inoculation of the AVP-treated seedlingts caused significant increase in the catalase activity at 1 DAI, 3 DAI and 5 DAI but this increased enzymatic activity also showed a gradual decline from 1 DAI to 5 DAI. B. spectabilis was more effective in increasing the level of the enzyme and phenolics than that of acetyl salicylic acid.
