Rapid PCR-based method for detection of canine parvovirus in dog faeces

Abstract

A polymerase chain reaction (PCR) based rapid detection test was established for detection of canine parvovirus (CPV) using primers designed from VP2 gene. The target DNA was extracted from infected cell culture supernatant and faecal samples using either spin column chromatography or direct heat treatment. PCR reaction was optimized to amplify 448 bp DNA sequence within VP2 gene. The amplified PCR product was characterized by restriction endonuclease analysis. The results of the present study show that sample preparation by direct heat treatment of clinical faecal samples can be used for rapid and reliable detection of CPV in referral diagnostic centres.
