A virus isolate was identified and characterized on sugarcane (var. CoLk 8102) showing systemic chlorotic stripes/streaks symptoms. Flexuous filamentous particles having size of 850x14 nm were observed under electron microscope in leaf dip preparation. The virus under study reacted strongly with polyclonal antiserum of sugarcane streak mosaic virus (SCSMV) in serological tests (DAC-ELISA and EBIA) and uniform decoration was also observed with the antiserum of SCSMV in ISEM test. However, it failed to react with potyvirus group specific antisera and with antisera of several well-characterized potyviruses. The molecular weight of coat protein was 39 kDa. Comparison of the 3’ terminal nucleotide sequence of the RNA genome of the virus revealed 93% sequence identity in the coat protein coding region with the recently described Sugarcane streak mosaic virus-AP isolate from south India, Pakistan and USA. This is the first report on the molecular characterization of a virus causing mosaic diseases of sugarcane in North India. The results demonstrate that the virus is a strain of Sugarcane streak mosaic virus of the family Potyviridae.
