Viruses are the most threatening pathogens of commercial cultivation potato crop, of which Potato virus Y (PVY) one of the imposed viruses. PVY is a ssRNA virus transmitted by aphids and mechanical means in potato transgenic potato through Coat Protein (CP) mediated resistance is a viable option for this virus. We cloned the CP gene from kolar isolate of PVY to eventually develop a transgenic potato. The virus RNA was extracted from tobacco leaf infected with PVY. The first strand of cDNA was synthesized from viral RNA template primed with synthetic oligo-nucleotide primer. A DNA fragment with 0.81 kb was obtained using gene specific primers. The sequencing of the fragment revealed that the gene possessed typical features including DAG signature. Sequence homology with other available CP sequences of PVY was studied. Results on expression homology features and expression in potato will be discussed.
