Abstract

Pigeonpea sterility mosaic virus (PPSMV) that causes sterility mosaic disease in pigeonpea is endemic in the subcontinent. Various isolates of this virus occur in the subcontinent. PPSMV isolates from Bangalore (B), Coimbatore (C), Dharwad (D), Gulbarga (G) and Patancheru (P)] and Varanasi (V), India are being studied. Based on the isolate reaction on a set of differential genotypes, these were grouped as ‘P type [P, D, G] and B types [B, C, V]’. The polyclonal antibodies raised to PPSMV isolate P can detect all the six isolates. Work is in progress to develop diagnostics tools for differential diagnosis of various PPSMV isolates. Polyclonal antibodies have been produced to C isolate, which differential react with homologous and heterogonous isolates. Using the single nucleotide differences found in the RNA-5 sequence of C and P isolates, oligonucleotide primers were designed for differential detection of C and P isolates by RT-PCR or by restriction enzyme analysis. Details of the progress made in differentiating P and C isolates will be presented.
