Abstract

Classical swine fever virus (CSFV) is a highly contagious and economically devastating disease of pigs. Accurate diagnosis of the disease enables to depict epidemiology and helps in formulating strategy for control of CSF outbreaks. Various antibody-based and molecular techniques are used for detecting CSFV in tissue samples. In the present study RT-PCR and nested PCR were used to amplify E2 region of the CSFV genome. Sandwich ELISA and direct FAT were performed to demonstrate viral antigen in the tissue samples. A total of 89 tissue samples comprising of tonsil (12), mesenteric lymph node (20), spleen (43) and kidney (14) were collected from pigs affected with CSF. In PCR 69.7% samples were found positive, which was followed by FAT (44.9%) and ELISA (41.6%). Tonsilar tissues were found more suitable (90.0%) in detecting CSFV by all three tests. In spleen tissue antibody based techniques could detect viral antigen in 25.0% samples but 76.0% samples were found positive by PCR. The molecular diagnostic technique is found most sensitive in detecting CSFV in tissue samples.
