Isolation of rotavirus in MA 104 cell line from diarrhoeic piglets of Assam

Abstract

Rotavirus (RV) has emerged as one of the important causes of piglet diarrhoea. In the present study, rotavirus was detected in faeces of infected pigs by ELISA and PAGE. Rotavirus was isolated from faecal samples using MA 104 cell culture. ELISA could detect the RV antigen in slightly higher numbers (29.35%) than that of PAGE (24.84%). A total of 18 samples were passaged in MA 104 cell culture up to the 8th passage. Out of which in 16 samples RV could be detected after completion of 8th passage. Interestingly virus isolated from two samples showed CPE characterized by elongation and rounding of cells. But remaining isolates were non-CPE producing. The propagated virus was detected by FAT, ELISA and PAGE.
