Abstract

Citrus mosaic disease is a serious disease of citrus in southern parts of India. The disease is caused by Citrus mosaic virus belonging to genus Badnavirus. CMBV is found associated with sathgudi sweet orange, acid lime, Pummelo and Rangpur lime. The disease produces mild to bright yellow mottling symptoms in different species of citrus. In order to determine the distribution of CMBV in the infected citrus plants, PCR detection was used to determine its presence and relative distribution in leaf, bark and bud tissues of infected sweet orange, Pummelo and Rangpur lime. The DNA template was prepared by simplified NCM based protocol and was used in standardized PCR reaction. Amplification of CMBV was observed in all the tested tissues. However, the dilution studies indicated that,the concentration of CMBV was maximum in bark followed by leaves and buds. It is concluded that bark can be a better source material for PCR detection of CMBV in infected citrus plant.
