The present investigation was undertaken to determine the prevalence of rotavirus in diarrheal & non-diarrheal feacalsamples of bovine calves. In all 38 faecal samples were collected from healthy as well as diarrhoeic calves of one day to two months of age from cattle breeding farm maintained at Nagpur Veterinary College, Nagpur. The faecal samples were suspended in PBS at a concentration of 10 percent. The ds RNA from the fecal samples was extracted using SDS and phenol chloroform mixture technique. RNA-PAGE was performed in vertical slab gel electrophoresis at 8 volt/cm rate. The gel was stained by silver nitrate for visualization of RNA bands. Out of 38 diarrhoeic fecal samples screened none was found to be positive by PAGE which suggests absence of rota virus infection. From the present study it is evident that the electropherotyping could be used as a routine technique for detecting as well as comparing rotaviruses. Failure to detect viruses from field sample should in no way, underestimate the importance of this agent in causing neonatal diarrhea as its significance has been well documented.
