Abstract

Virus diseases have been of great concern for floriculture industry in Himachal Pradesh, causing significant yield loss in quality and quantity. Bean Yellow Mosaic Virus (BYMV) is one of the major viruses causing infection to this crop in HP. During these investigations, gladioli showing virus like symptoms were induced, causal virus was isolated, sequenced and then the nucleotide and amino acid sequences were analyzed using bioinformatics tools. Gladiolus plants growing in floriculture fields and greenhouse were infected by a viral disease showing symptoms of mosaic and streak on the leaves and colour breaking in flowers. Out of total RNA isolated, BYMV CP gene mRNA was converted in cDNA using CP gene specific primers and further PCR results gave an amplicon of ∼850bp. Plasmid was isolated from transformed E. coli bacterial cultures and restricted with Eco RI. All the bacterial cultures (1, 2&3) showed the presence of gene insert of exact size i.e. ∼850bp. Sequence comparison showed that Solan and Palampur BYMV isolates infecting gladiolus are similar, confirming their same parentage. Pair wise sequence alignment showed that Japan isolates are showing maximum variability with Solan isolate while multiple sequence alignment showed that the hinder part of all the sequences at nucleotide as well as amino acid level are similar, confirming the presence of a conserved region. Phylogenetic analysis clearly showed that the Japan isolates cluster together and showing maximum variation from Solan isolate of BYMV. Phylogrames have been extensively used to study stainal relationships, and it has been cited that stainal variation observed within the stains of BYMV.
