Abstract

Common bean (Phaseolus vulgaris L.) locally known as “Rajmash” is affected severely by common mosaic disease caused by Bean common mosaic virus (BCMV), a positive stranded RNA virus belonging to family Potyviridae and known to possess high degree of pathogenic variability throughout the world. In H. P., four strains have been reported to exist on common bean. NL-1n strain is most prevalent so an attempt was made to characterize 3’ terminal region of its genome. Viral RNA was isolated from infected leaves by Trizol reagent. PCR amplification of cDNA synthesized by oligodT using BCMV specific primers generated an amplicon of ∼1300bp, which was cloned in pGEMT-Easy vector and custom sequenced. Sequence results after blast analysis showed the product comprising of partial NIb (220bp), complete coat protein region (860bp) and 3’ untranslated region (208bp). The sequence has been submitted in NCBI GenBank vide accession no. EU 713858. Multiple alignment of the 3’ terminal region of the test strain by ClustalW program showed maximum homology with the NL-1 type strain of BCMV. This constitutes the first report of sequence of necrotic isolate of BCMV from India.
