Abstract

A virus causing chlorosis and vein clearing on carrot (Daucus carota) were observed in various locations with 33% disease incidence during November-December in 2007 in Aligarh, India. The causal agent produced chlorotic local lesion after 5 days and systemic were 20 days post inoculation on Chenopodium amaranticolor and Nicotiana tabacum cv Samsun NN. Leaf dip preparations revealed the presence of 28nm spherical core particles, supporting the association of cucumovirus with disease. The present virus has been identified as an isolate of Cucumber mosaic virus subgroup II on the basis of host range studies, transmission by aphids in non-persistent manner, reverse transcription polymerase chain reaction RT-PCR, enzyme linked immunosorbent assay (ELISA) with Cucumber mosaic virus specific polyclonal antiserum along with alkaline phosphates-linked antibodies. Virus transmission efficiency by mechanical inoculation was 75 to 80% and by Myzus persicae and Aphis gossypii were 30 to 40%. The host range included members of the Apiaceae, Chenopodiaceae, Cucurbitaceae, Leguminosae and Solanaceae families. A complete coat protein gene was amplified with CMV specific primers, (choi et al., 1999), ∼950 bp band contained from infected Daucus carota was cloned and get sequenced. In silico analysis of sequence data showed ~657 bp long capsid protein open reading frame translating 218 amino acid which was submitted to GenBank (Acc. EU600214). Highest 96% CP nucleotide identity was obtained with Tagetes strains (AM396983) of CMV subgroup II, when analyzed by BLAST analysis. During Phylogenetic analysis, CMV isolate under study distinctly clade with subgroup II reported from India and abroad.
