Pathology of female genitalia of goats.

The present investigation was planned to study the incidence of different pathological conditions affecting female genitalia of goats and identification of important aetiological agents involved in causation of reproductive diseases in goats. During the period of study a total of 660 samples were collected irrespective of different age group and breeds from various abattoirs in U.P, Haryana and Delhi state and from CIRG postmortem facility. An overall incidence of pathological conditions was 23.32% (154/660), wherein the incidence of affected uterus was found to be highest (12.42%), followed by ovary (6.06%), cervix (2.57%), fallopian tubes (1.21%) and vagina (1.06%). The lesions that resulted into infertility or sterility were found to be 12.27%. Endometritis (37/154; 24.02%) was the most common pathological lesion noticed followed by ovarian cyst (22/154; 14.28%). Other conditions like congenital anomalies (3/154; 1.94%), hydrosalpinx (2/154; 1.29%), cervicitis (10/154; 6.4%), vaginitis (3/154; 1.94%) etc. were observed. The suspected samples (n=76) based on gross and microscopic lesions like uterine caruncular tips showing petechial haemorrhages, presence of remnant of placenta, endometritis, cervicitis etc were subjected to PCR test, which revealed the occurrence of Campylobacter, Brucella and Chlamydia infection to be 5.03%, 1.81% and 0.45%, respectively in various reproductive disease conditions in goats. Brucella melitensis antigen was localised in formalin-fixed paraffin-embedded tissue sections of uterus employing immunohistochemistry, which evinced cytoplasmic immunolabelling of macrophages indicating the presence of organism and role played by macrophages in the infection. A critical analysis of gene expression profile of an endometritic uterus vis-à-vis normal uterus showed that 4238 genes were up-regulated and 4328 genes were down-regulated. Various genes including CCR3, IL8, IL15, PTGS2, IFNAR2 and IL2RA with most of them involved in Pro-inflammatory chemokine and cytokine signalling pathways were significantly upregulated. On the other hand, oxidative stress responses mediated through a series of related genes viz., TP53, MYC, IL18 and TXN showed a significant (P<1.00) down regulated transcriptional response in inflamed uterus compared to the normal.
