Induced toxicity of zinc sulphate in cockerels

India The present experiment was carried out to assess the toxico-pathological effects of zinc sulphate in cockerels. For carrying out the experiment a total of 80 WLH male birds (one month old) were randomly divided into four groups (C, G1, G2 and G3) of 20 birds each. Zinc sulphate was incorporated in drinking water @ 15000 ppm, 25000ppm and 35000ppm to group G1, G2 and G3 respectively for a period of 12 weeks. The birds of group C served as a control. Four slaughters have been carried out in this study on 20, 40, 60, and 90^th day of study. At every slaughter blood and serum samples were collected to assess the haematological, biochemical, antioxidative and immunological parameters. From each group a total of five birds were sacrificed at every slaughter to study the pathomorphology of organs and to collect the tissues for assessment of antioxidative status. The average body weights of experimental birds were measured after every 14 days interval revealed a significant average body weight reduction in zinc intoxicated birds after 28^th day of study. Effects of zinc sulphate on the haematological parameters were significantly decreased haemoglobin, PCV, TEC and TLC, however a significant increase in haemoglobin and PCV in group G2 during IV slaughter and G3 during III and IV slaughter were observed. A non significant variation in differential leucocyte count was observed in all the experimental birds. The results of biochemical parameters revealed a significant decrease in glucose and total cholesterol in all the experimental groups as compared to control. The levels of total protein, albumin and globulin were significantly decreased in group G1 from I to IV slaughter, while in group G2 and G3 a significant linear decrease in total protein, albumin and globulin were observed from I to III slaughter, while in IV slaughter a significant increase was observed in group G2. In group G3 a significant increase was observed in III and IV slaughter as compared to II slaughter. A significant increase in BUN, creatinine, ALT, AST and ALP levels were found in all the experimental birds as compared to control birds. Moreover, these values were in ascending order from control to G1, G2 and G3.The results of antioxidative parameters in RBCs and tissues revealed a significant increase in LPO level, while GSH was found significantly decreased in all the Zn intoxicated birds. A significant decrease in serum immunoglobulin and DTH response was observed in all the experimental intoxicated birds as compared to control. A significant variation in organ/body weight ratio was observed for liver, kidney, spleen and brain; however a non significant decrease in organ/body weight ratio for pancreas was observed in group G1 and G2. Grossly, congestion and atrophy of various organs were observed. Histopathological examination revealed gradual degenerative and inflammatory changes in liver, pancreas, kidney gizzard, bursa of Fabricius, spleen and intestine during all four stages ofslaughter in all Zn intoxicated groups with dose dependent progressive lesions. However lungs, brain and testes did not revealed any microscopic changes. From the present study, it may be concluded that excess zinc has adverse effects on the general health, body weight, haematobiochemical, oxidative and immune status of the birds and these ill effects are dependent on dose and duration of exposure of zinc.
