The expression of ê-casein is controlled by different regulatory elements which act through its promoter. Genomic DNA was isolated from buffalo white blood cells (WBCs). 5′-flanking region of êcasein gene was amplified using PCR and sequenced. The presence of highly conserved putative transcription factor binding sites viz. MGF/STAT5, AP-1, YY1 and C/EBP in 5’ flanking region of buffalo ê-casein were reported. Homologies of 96.5%, 93.5%, 91.7%, 68.3% and 57.2% were found with bovine, goat, sheep, camel and horse respectively. A phylogenetic tree constructed on the basis of sequence comparisons showed that buffalo is evolutionary more closely related to cattle with respect to this gene.
