Immunobiochemical characterization of 55 kDa fertility associated protein of Garole Sheep (Ovis aries) seminal plasma

Abstract

The objective of this study was to characterize a 55 kDa protein in Garole sheep (Ovis aries) seminal plasma previously shown to be prevalent in bulls and swine of high fertility. Seminal plasma was partially purified by gel filtration chromatography. 10% SDS-PAGE analysis for their polypeptide profile, revealed a prominent band of 55 kDa including two other minor bands of 69 kDa and 16 kDa. Hyper immune sera was raised in rabbit and immuno-dominant polypeptide was studied using DID, indirect ELISA and Western Blot analysis. Partially purified fraction containing 55kDa protein showed specific immuno reactivity in DID and indirect ELISA. This was further confirmed by the Western Blot analysis where immuno-reactive polypeptide band of 55 kDa was found. These findings indicate that Garole sheep (Ovis aries) seminal plasma contain this 55 kDa protein, also evident in bovidae and suidae species, is a prominent immuno-dominant protein and may have pivotal role in fertility of this breed.
