A gene encoding mtc28, immuno dominant proline-rich 28 kDa protein, was characterized in the Indian field strain of Mycobacterium bovis (Rv3/86). Recombinant plasmid named pET28b-mtc28 was constructed via inserting the mtc28 gene into the pET vector and sequenced. The nucleotide sequence and its deduced amino acid sequence were compared with the published sequences of Mycobacterium tuberculosis complex (MTBC). The sequence of the mtc28 gene shared 98.8 to 99.4% nucleotide homology and 98.2 to 98.9% deduced protein sequence homology for all studied genes with published MTBC. There was a very low amino acid sequence homology with 70.9% with M. ulcerans and 51.8% with M. smegmatis.
