Effect of dimethyl sulphoxide (DMSO) on acrosomal intactness in buck semen

Abstract

Semen from six Osmanabadi and six Sirohi bucks was diluted using Tris Egg yolk DMSO (egg yolk 5% and DMSO 5%) dilutor (TYD). Semen was evaluated for acrsomal intactness by Giemsa staining. The acrosome intactness of spermatozoa has significantly reduced from fresh semen to pre-freezing in Osmanabadi buck semen but it was not significantly reduced in Sirohi buck semen. In semen of both breeds there was no significant reduction in acrosome intact sperm percent from pre freezing to post freezing (24 and 72 hours) stage. This indicates that Dimethyl sulfoxide (DMSO) was effective in maintaining acrosome intactness of Osmanbadi and Sirohi buck spermatozoa on cryopreservation of semen.
