Prolactin (PRL) plays an important role in promoting mammalian mammary gland development, and milk production during lactation. In the present study, association of polymorphic variants of PRL gene at exon 3 region with milk production traits in Frieswal cattle was investigated. Genomic DNA was isolated from blood samples of 54 Frieswal cattle. A 156 bp PRL gene exon-3 segment was amplified by PCR using bovine specific primers. Restriction Fragment Length Polymorphism (RFLP) using RsaI restriction enzyme revealed two alleles A and B with 0.630 and 0.370 allele frequencies, respectively. Genotypic frequencies were 0.315 for AA, 0.629 for AB and 0.056 for BB genotypes in Frieswal cattle. The result showed that the population was not in Hardy-Weinberg equilibrium. The animals with BB genotype had higher lactation length, where as AB genotype had higher lactation yield. Shorter service period was found to be associated with AA genotype. From the study, it may be concluded that the prolactin gene is considerable marker for genetic selection of dairy traits in Frieswal cattle.
