Effect of disaccharides in semen extender on acrosomal intigrity, acrosin content and penetrating ability of bovine spermatozoa*

Abstract

The present experiment was conducted to examine effect of addition of disaccharide sugars (lactose or sucrose) in tris-egg yolk-glucose/fructose extenders during cryopreservation process of bovine semen. Significantly higher (P < 0.01) percent counts of pre-freeze and frozen-thawed spermatozoa with intact acrosome were observed by initially providing lactose or sucrose in both the extenders, except in lactose added tris-egg yolk-glucose extender. Significantly greater acrosin activity in pre-freeze and frozen-thawed spermatozoa was noticed by presence of sucrose (P < 0.01) in both the extenders and presence of lactose (P < 0.05) at extension and (P < 0.01) after cryopreservation in tris-egg yolk glucose extender. Significant (P < 0.01) increase in hamster egg penetration rate and index was noted for frozen thawed semen in lactose or sucrose added both the extenders. The presence of glucose in extender acted complementary with lactose or sucrose in retention of acrosomal integrity and acrosin content, effecting significantly (P < 0.01) higher hamster egg penetration rate for frozen thawed bovine spermatozoa.
