Standardisation of high performance liquid chromatographic technique for detection of gentamicin residues from animal tissues

Abstract

High performance liquid chromatographic (HPLC) technique employing acetonitrile:water as mobile phase, C18 column and fluorescence detector at 260 nm wavelength for excitation and 315 nm wavelength for emission was standardised for detection and quantitation of gentamicin residues. Muscle/organ samples were subjected to solid phase extraction, clean-up procedure and derivatization prior to chromatographic analysis. Gentamicin was eluted in 4 components viz. C1, C1a C2a and C2 within 15 minutes. Recoveries ranged between 41.89–48.66% in muscle, 45.23–61.21% in liver and 39.80–44.61% in kidney samples fortified with gentamicin at 0.05 to 1.4 ppm levels with detection limit 0.05 ppm.
