Cytogenetic investigation was undertaken in Red Kandhari cattle by short-term lymphocyte culture technique to compare and evaluate in terms of mitotic drive and mitotic index for their suitability for culturing of Red Kandhari cattle lymphocytes for chromosomal studies. The media compared were TC 199 and RPMI 1640 for their efficacy in terms of mitotic drives and mitotic indices. Mean mitotic drives obtained with TC 199 and RPMI 1640 respectively were 56.98 ± 0.57 and 51.02 ± 0.26 per cent for Red Kandhari cattle lymphocytes. The corresponding mitotic indices were 1.21 ± 0.06 and 0.70 ± 0.02 per cent. The media differed significantly (p < 0.05) for both mitotic drives and mitotic indices for Red Kandhari cattle lymphocytes. The medium TC 199 was found to be superior to RPMI 1640 for culturing Red Kandhari cattle lymphocytes.
