Matrix metalloproteinases (MMPs) are a group of multifunctional enzymes involved in physiological processes; however, MMPs are also involved in pathological conditions, such as pulp and periodontal diseases. Aim: The aim was to detect the amount of MMP-9 released from HeLa epithelial cells as a measure of the inflammatory impact of different root end filling and sealant materials on the periapical and periodontal tissues.
Standardized disks for the dental materials tested in the study were prepared using a brass mold prior to its incubation with HeLa cells. The materials used were: Amalgam, composite, flowable composite, zinc oxide eugenol (ZOE) sealer, nano-ZOE sealer, and resin-modified glass ionomer. Results: Enzyme linked-immunosorbent assay-based determination of MMP-9 released into the culture media showed that the highest level of MMP-9 was measured with the flowable composite material, then with amalgam, followed by glass ionomer (Photac Fil). Then, the ZOE sealer came in the fourth level and followed by nano ZOE. The best results came with the Filtek composite material which produced similar results as the control group.
The Filtek composite biomaterial when used as retrograde filling material produces the least irritation of HeLa epithelial cells with least induction of inflammatory cytokine MMP-9.
