Toxicological evaluation of ampucare: acute oral toxicity in wistar rats

Abstract

Toxicological evaluation of ampucare was done on wistar rats of both sexes. Forty wistar rats weighing from 100 to 125 g, were divided in four groups, with 5 males and 5 females per each group. They were kept in laboratory's animal house and housed in standard polypropylene rat cages with stainless steel grill using paddy husk as the bedding material. Acclimatization was done 5 days prior to test in the test chamber. The test room was air-conditioned with temperature 25 ± 3°C, relative humidity 65 ± 5% and with 12 h artificial fluorescent light and 12 h dark. Animals were fed with standard rat pellet feed. Purified water was provided ad libitum. Group II, III and IV, were given ampucare at the dose of 1250, 1500 and 2000 mg/kg body weight respectively. Group I received distilled water alone. Dose volume was maintained at 10 ml/kg body weight. All the animals were monitored individually for 14 days after the administration of ampucare. Body weight, mortality, toxic signs, autopsy and necropsy were the parameters and Finney's probit analysis was employed to calculate the LD50. The changes in body weight of experimental rat were compared with that of the control using student's t-test. The results of overall study showed the LD50 value of oral toxicity of ampucare to be above 2000 mg/kg body weight in wistar rats of both sexes.
