Standardization of media supplements for in vitro propagation of orchid

Abstract

Experiment was carried out to standardize media supplements for shoot proliferation, for root initiation and development of orchid (Dendrobium formosum) through in vitro propagation. The result revealed that MS medium fortified with 4.0 mg/L BAP, 1.0 mg/L IAA and 0.1 mg/L GA3 was found to be best for production of more number of protocorm-like bodies (PLBs) or explants (7.13 and 9.27) and enhancement of length of PLBs (2.00 and 2.03 cm) and length of the leaf (1.16 and 1.46) after an interval of 30 days of culture and sub-culture respectively. MS medim supplemented with 3.0 mg/L BAP, 1.5 mg/L IAA and 0.1 mg/L NAA was found to be most potent combination for production of more number of shoots/explants (4.2 and 8.0), more number of leaves/shoot mass (9.48 and 11.48) and longer leaves (2.2 and 2.36 cm) after 60 and 90 days of subcultures respectively. As regards to root initiation and formation MS medium fortified with 1.0 mg/L IBA and 1.0mg/L NAA was found to be best for obtaining higher number of roots (3.5) and longer roots after 60 days of culture.
