Protein profile was resolved using SDS-PAGE with an objective to correlate it with sequential stages of differentiation in callus cultures of Cardiospermum halicacabum. The callus was initiated from leaf explant on MS basal medium supplemented with 1.0 mg/l each of 2, 4-D and Kn. The cultures were incubated at 26±2ºC under a photoperiod of 16 h light (2000 lux) and 8 h of darkness. The callus was transferred to media supplemented with BAP (0.5 mg/l) + NAA (0.3 mg/l) and BAP (0.5 mg/l) + NAA(0.1 mg/l) after 4 week of inoculation for differentiation of root and shoot, respectively. Root differentiating calli revealed a steady increase in the protein content upto12 days after inoculation while a sharp increase in protein content was evident after 8 days of inoculation in shoot differentiating calli. Protein profile studies revealed appearance of four common bends (51.29, 46.77, 33.8 and 30.2 kDa) during differentiation of root as well as shoot. In addition, one root specific (85.1 kDa) and two shoot specific (102.3 and 36.31 kDa) protein bands were evident.
