IBT/015 Comparative Evaluation of HI Test and c-ELISA in Diagnosis of Peste Des Petits Ruminant in Sheep and Goats

Abstract

A total of 432 serum samples collected from 216 sheep and 216 goats were tested for Peste Des Petits Ruminant (PPR) antibodies by competitive-ELISA (c-ELISA) and haemagglutination inhibition (HI) test. Out of 432 samples tested 125 and 110 samples were found positive by c-ELISA and HI test, respectively. 14 samples negative by c-ELISA were detected positive by HI test and 29 samples tested negative by HI yielded positive results by c-ELISA. Whereas both the tests reported 96 and 293 samples as positive and negative, respectively. The relative sensitivity and specificity of HI test compared with c-ELISA was 76.8% and 95.40%, respectively, with positive predictive value of 0.873 and negative predictive value of 0.910, Whereas c-ELISA has relative sensitivity and specificity of 87.30% and 91%, respectively, when compared with HI test with positive predictive value of 0.786 and negative predictive value of 0.954. The overall agreement between the two tests was 90%, which was further supported by kappa value of 0.749, suggesting a substantial agreement between the two tests. Species wise analysis revealed that HI was more sensitive 78.70% in goats than in sheep 75.00%, but was less specific in goats 94.80% than in sheep 96.1% in diagnosing of PPR antibodies in serum samples.
