Different enzyme labels are used with different chromogenic substrate mixtures for developing ELISA. Horse-radish peroxidase enzyme alongwith O-phenylenediamine (OPD) and hydrogen peroxide (H2O2) is one of the commonly used systems. Use of solution of H2O2 poses many problems including loss in activity with time. In this study, sodium perborate was evaluated for its efficiency as substitute for H2O2. Anti Pasteurella multocida outer membrane protein antibody titres for 92 cattle sera samples were calculated by end pomt indirect ELISA using the substrate solution with sodium perborate or solution of H2O2. The antibody titres obtained by using these two substrate systems were found comparable. Therefore, sodium perborate could be used in the chromogen-substrate buffer in place of solution of H2O2 without having any ill or negative effect on performance of ELISA.
