Peripheral blood mononuclear cells were separated from the blood of Nilgai (Boselaphus tragocamelus) using Histopaque density gradient centrifugation. The cells were stimulated with ConA at 37°C in a humidified atmosphere containing 5% CO2. After 20 h incubation, the total RNA was isolated from the stimulated cells, cDNA was synthesized and the full gene for Interkeukin-18 (IL-18) was amplified using gene-specific primers. The amplified IL-18 gene was cloned in T/A cloning vector and the recombinant plasmids were characterized by PCR and Pst I restriction digestion. The nucleotide sequences of IL-18 inseret were determined. The sequence analysis revealed an open reading frame of 582 bp corresponding to IL-18 gene. This sequence was submitted to Genbank (accession no. AY842499).
