A study was conducted to compare the in vitro propagation response in Single and Double petalled tuberose cultivars namely, Prajwal and Vaibhav, respectively. Initially aseptic cultures were established using 0.1 per cent of carbendazim and mancozeb and 200 mg l−18-HQC for 4h as pre-treatment and (1.0%) of HgCl2 and NaOCI for 8 min. as surface sterilent. It was demonstrated that the MS medium supplemented with 4 mg l−1 BAP enhanced shoot multiplication in both Single (3.3) and Double (2.5) cultivars using bulb scale. Maximum number of roots (21.63) were observed on half-strength MS medium +1 mg l−1 IBA which also gave the highest rooting (91.6%). The in vitro raised miiro shoots were successfully yaclimatized in glass jar with polypropylene cap and were transferred to glasshouse conditions.
