Abstract

Protocol for in vitro rapid clonal multiplication of Dendrobium cv. Sonia was investigated. Maximum establishment of cultures of offest segments occurred on Murashige and Skoog (MS) medium containing 2 µM NAA + 5 µM BAP or 2 µM NAA + 10 µM KIN. MS medium supplemented with 2 µM NAA + 6 µM BAP + 3.4 µM paclobutrazol (PP-333) was optimum for shoot multiplication. Abundant protocom like bodies (Plb's) were formed in MS medium containing 10 µM each of NAA and BAP; these were excellent for further multiplication of shoots. NAA (6 or 8 µM) or IBA (4 or 6 µM) in MS medium was suitable for the rooting of in vitro multiplied shoots.
