Association of ‘Ca. Phytoplasma asteris ’(16SrI group) with little leaf and witches ’broom symptoms of Catharanthus roseus

Abstract

During survey of Gorakhpur area in August 2014, Cathranthus roseus plants showing little leaf and witches ’broom symptoms were analyzed for phytoplasma presence through nested PCR assays with universal phytoplasma primers P1/P6 and R16F2n/R16R2. 1.2kb amplicon was observed in symptomatic and positive control sample. The amplified products were eluted and directly sequenced. Aligned sequences were submitted in the GenBank with the accession number KX894734. BLASTn search analysis of 16S rDNA sequences of Catharanthus phytoplasma Gorakhpur isolate showed maximum identity with identified strains of 'Ca P asteris ’which was further supported by phylogenetic analysis.
