Precocious Shoot Proliferation and Microcorm Production in Gladiolus Through Tissue Culture

Abstract

In vitro studies were conducted for rapid clonal multiplication and microcorm production in gladiolus cvs Eurovision and Wine & Roses. Cormels were surface sterilized with mercuric chloride 0.1% for 8–10 minutes. MS medium supplemented with 2 mM NAA, 10–20 mM BAP and 2 mM GA3 was optimal for the culture establishment of both cultivars. Early sprouting as well faster increase in sprout length occurred with the addition of GA3 in the establishment medium. Profuse shoot proliferation was obtained on GMM-7(MS+2 mM KIN) in cv. Wine & Roses while it was optimal on GMM-8 (MS + 4 mM KIN) in case of cv. Eurovision. Half strength MS medium (liquid) containing 10 mM IB A, 5 mM KIN and 10 mM PBZ was most satisfactory for profuse microcorm production when kept under dark environment.
