Regeneration response of in vitro derived leaf explants of Gerbera jamesonii

Abstract

The response of in vitro, leaf explants of Gerbera jamesonii to callusing and regeneration was studied in full and half strength Murashige and Skoog's (1962) media supplemented with different concentrations of 2,4-D and BAP and IAA. Leaf explants cultured on MS medium fortified with low level of 2,4-D (0.5 mg l−1) and high concentration of BAP (3.0 mg l−1) produced creamy or creamy yellow callus within 16 days. But leaf explants cultured on MS medium supplemented with different concentrations of IAA and BAP produced green or greenish white callus. Direct plant regeneration was maximum from in vitro petiole tips of 2nd, 3rd or 4th leaf cultured on MS medium fortified with 0.25 mg l−1 IAA + 1 mg l−1 BAP.
