The influence of plant growth substances and media on protocorm (Plbs) proliferation, differentiation and growth of Cymbidium Golden Girl was assessed. Maximum number of Plbs (6.67) was obtained on Murashige and Skoog media containing lg/l AC with 0.25 mg/l, followed by MS with 0.25 mg/l BAP and 0.1 mg/l NAA (6.33). Maximum plant growth (186.67 mm) with elongated root (88 mm) was obtained in Nitsch media in combination with 0.5 mg/l Triacontanol and 0.5 mg/l PBZ. Leaf length was also more I (114.33mm) in Nitsch with 0.5mg/l each of Tria and PBZ, followed by 100.33 mm on the I same media incorporated with 0.25mg/l each of Tria and PBZ. Rooted plantlets having 4-5 well developed leaves pre hardened in half strength MS salts containing 0.5 mg/l each of PBZ and Triacontanol recorded 88.3% survival ex vitro.
