In the era of advancement and modern technology, PCR technology has substantiated its valuable portrayal and become the witness of tremendous changes. This technology is significant to observe the changes and its accuracy in DNA profiling of forensic cases. DNA fingerprinting is considered perpetual, ubiquitous and exclusive for the establishment of individuality. It is based on the polymorphism in DNA sequence. Careful examination of trace evidences along with STR technology can be a milestone, which can change the favour of any cases. In this present case, DNA technology helped to whack the perpetrator of the murder of a young girl. The physical exploration had no sign of any fresh sexual assault, yet the vaginal swab was recovered. In any sexual assault, vaginal swab epitomizes the distinct ratio of male and female DNA because of limited number of sperms and high concentration of female cell. A forensic scientist always uses this ratio to determine the identity of culprit by differential extraction. Due to the limited amount of vaginal swab, scientists prefer the direct examination rather than differential extraction. In this case, direct examination helped to extract out enough information/profile of culprit, that was sufficient to nab him and put behind the bar.
