Diagnosis of Citrus Yellow Mosaic Virus by PCR and Elisa in Sweet Orange (Citrus Sinensis Osbeck)

Abstract

Citrus Yellow Mosaic Virus (CYMV) is one of the major viral disease of sweet orange and responsible to produce lower fruit yield but also produce less juice and ascorbic acid, productive life of the trees is also reduced. It was reported from Andhra Pradesh (AP) and later form other Citrus growing areas in the country. A preliminary survey on prevalence of CYMV associated with Sweet orange was carried out in three major Sweet orange growing districts of A. P and.the.results revealed that a high incidence of 20.8% was recorded in Nellore district followed by Nalgonda (16, 5%) and Kadapa (7.3%). The DNA extracted from different plant parts of Sweet orange i.e. Leaf, Twig bark, Fruit-rind, Fruit-rag and Fruit-juice by addition of Sodium sulphite to Tris-EDTA method were subjected to PGR amplification. An amplification product of 710 bp was observed in leaf, twig bark, fruit-rind and fruit-juice. No amplification was observed in fruit-rag. A bright amplification was observed in case of leaf and twig bark compared to fruit-rind and fruit-juice. It can be concluded that leaf and twig bark samples will be of more suitable for detection 6f this virus. Serologically detected CYMV from different plant parts of sweet orange by DAC-ELISA at 1:1000 dilution.
