Sepsis Differentially Regulates Expression of CYP450 Isoforms in Liver and Kidney of Mice

Abstract

CYP450 enzymes are well known for xenobiotic biotransformation and their plethora of effects ranging from cardio-protection, vasodilation, pro-angiogenesis, pro-inflammation, and anti-inflammation. The present study was designed to evaluate the effect of sepsis on the expression of CYP2 isoforms viz. Cyp2c29 and Cyp2j9. Sepsis was induced by the caecal ligation and puncture (CLP) method. A significant down-regulation of Cyp2c29 and upregulation of Cyp2j9 isoforms in the liver of septic mice were found. On the contrary, non-significant down-expression of Cyp2j9 was found in the kidneys of septic mice. Liver and kidney histopathology showed marked cellular changes accompanied by cellular swelling, degeneration or necrotic changes, and inflammatory changes in the hepatocytes and renal tissue. Taken together, it may be inferred that organ-specific regulation of Cyp2 isoform in metabolic (liver) and excretory (kidney) organs during sepsis may differentially affect the disposition of clinically used drugs that are metabolised by these enzyme isoforms.
