Pharmacokinetics of danofloxacin after intravenous administration in rabbits

Abstract

The present study was conducted to investigate the pharmacokinetic properties of danofloxacin in rabbits. Plasma concentrations of danofloxacin were determined by ultra-high performance liquid chromatography (UHPLC). Pharmacokinetic parameters were derived from plasma concentrations and time data using PK Solver 2.0 software. After a single intravenous injection of danofloxacin at 5.0 mg/kg body weight, the elimination rate constant (2) values ranged from 0.14 to 0.18 1/h, with a mean value of 0.16 1/h. The calculated mean value of the corresponding half-life (t1/22) was 4.31 h, with a range from 3.94 to 4.96 h. The mean value for the area under the curve (AUC) of plasma concentration of danofloxacin versus time was 6.09 μg.h/mL, and the mean value for the area under the first moment of plasma concentration (AUMC) was 22.44 1/4g.h2 /mL. The mean residence time (MRT) averaged 3.68 hours. The mean apparent volume of distribution (Vd(area)) and steady-state volume of distribution (Vd(ss)) were 5.12 and 3.03 L/kg, respectively. The mean value of total body clearance (ClB) was 0.82 L/kg/h.
