Shoot bud induction in non-differentiating hypocotyl explants by physiological manipulations in chickpea (Cicer arietinum L.)

Abstract

In the present study attempts were made to induce differentiation in hypocotyl (H) explants, which were observed to be non-differentiating in in-vitro culture. Several physiological factors influ. enced bud induction. Sucrose concentration in the medium was increased to 6% to obtain optimum bud induction. Similarly supplementing the media with amino acid, cysteine (50 mg I−1)and an ethylene action inhibitor AgNO3 (25 μM, enhanced shoot bud induction. Of the two treatments silver nitrate (AgNO3) treatment was found to produce more bud differentiation. Orientation of explant in in-vitro culture seems to play an important role in inducing shoot bud differentiation in hypocotyl explants..
