Stability is an important preanalytical parameter which has to be determined prior to its LC–MS/MS analysis. The instability of some analytes is readily predictable because of the presence of certain chemically or biologically labile moieties, such as ester, thiol, and catechol and others, in the molecules. The factors of instability of molecules should be assayed prior to analysis to avoid misinterpretation of analyte concentration during quantitative analysis. Regulatory always demands for certain documents stating cause of instability and impact on analysis based on proper scientific facts and figures with the conclusion. This review article comprises the facts and figures with support of in-house experiments and result to understand the concepts of instability of molecules during bioanalysis of drugs.
