Correlation of genomic diversity with bioavailability of drugs

Abstract

Genetic variants in the pharmacokinetic (PK) mechanism are the main underlying factors affecting the drug response and variability. In this review we examine various proteins that are important to the absorption, distribution and excretion of a large number of drugs in clinical use. We provide our assessment of the potential impact of known polymorphisms in such transporters, enzymes and discuss whether there is sufficient evidence to incorporate these polymorphisms in drug discovery and drug development process. Every volunteer and patient will have their own genetic constituents and genetic makeup, alteration of genetic constituents also alters the protein fashion and its bioactivity significantly. This review will compile the case studies from relevant noble research and review articles to attain the collective efforts in common platform and enlight the genetic facts of drug variability.
