Validation of RP-HPLC method for analysis of amaroswerin and amarogentin in Swertia chirayita (Roxb.) H. Karst.

Abstract

In the present study, RP-HPLC method was developed for the estimation of amaroswerin and amarogentin contents in Swertia chirayita. The developed method was found to be simple, accurate and precise. Separation was achieved with methanol and water in isocratic grade at 235 nm wavelength with good peaks of amaroswerin and amarogentin. Linearity was found in the calibration curve with correlation coefficient of 0.999 where concentration ranged from 5.93 µg/ml to 190 µg/ml. Limit of detection and limit of quantitation were found to be 0.197 and 0.593 for amaroswerin and 0.794 and 0.296 for amarogentin, respectively. Accuracy was determined with overall recovery of 101.09% for amaroswerin and of 102.30% for amarogentin. RSD value for inter-day and intra-day repeatability assay of both compounds under study was lesser than 3.
